hsa-mir-99b_Rep_s

The hsa-mir-99b_Rep_s (BBa_K5086100) is a composite element formed by combining hsa-mir-99b_Rep (BBa_K5086005) and Universalness_h (BBa_K5086009). In which the 5' end of hsa-mir-99b_Rep is complementarily paired and linked to the Universalness_h chain, and the remaining sequence of the 3' end of hsa-mir-99b_Rep_s in hsa-mir-99b_Rep_s can be complementary to the target hsa-mir-99b, which can be complementary to the target hsa-mir-99b. Therefore, it can specifically bind to the target to recognize the input signal. DNA polymerase polymerizes DNA from the 3' end of the target hsa-mir-99b in the 5'-3' direction, and in the process of DNA polymerization by Bst DNA polymerase, the Universalness_h strand is replaced by the newly synthesized daughter strand and released. Through this process, the target signal is converted to the Universalness_h strand, which continues to undergo subsequent strand replacement reactions to produce a fluorescent report. (The significance of this chain replacement step is that it facilitates the subsequent self-enhancement, and the DDSD probe shelf cannot be synthesized without the replacement of the Universalness_h chain.)
